User Manual (Use Process of Carlsberg flask)

1. Open the sealing wrench, mobilize the sealing pressure block, and open the top cover of the carlsberg flask.

2. Add about 80% of the total capacity of wort (medium,) to the cleaned Carlsberg flask, and seal the top cover of the Carlsberg flask

3. Heat the Carlsberg flask in an electric stove, steam pot or other heater to sterilize the wort (medium), and the boiling time is 30-40 minutes (steam sterilization can also be used, and it needs to be calculated separately based on temperature, pressure, etc.) The amount of wort and steam).

4. After sterilization, place the Carlsberg flask in a refrigerator or cold space to cool to room temperature naturally.

The aseptic breathing valve (and filter), sampling valve, safety valve, etc. should be installed before the wort is sterilized so that the steam discharged from the tank can be sterilized during the wort sterilization.

5. Oxygenate the wort cooled to room temperature. The oxygenation process should be carried out slowly. The wort will produce a lot of foam when placed, and the oxygenation time should be controlled at about 10 minutes.

6. After the completion of wort oxygenation, insert the original strain into the Carlsberg flask under aseptic conditions.

7. Carry out yeast aerobic culture at the specified temperature and time.

8. After the yeast cultivation is completed, pass in sterile air to press out the yeast to the yeast expansion tank. After the yeast is fully discharged, it is best to continue oxygenating for 1-2 minutes to ensure that the inoculation process is completed. It is best to close the inoculation container and the valve of Carlsberg flask stops the gas supply.

9. All connecting pipes, valves, interfaces, etc. must be thoroughly sterilized before inoculation.

10. The oxygen pressure does not exceed 0.15Mpa.

11. After the Carlsberg flask is used, it should be disassembled and washed in time, and generally cleaned manually (pipe cleaning fluid can be used).

Check the filter, cover seal ring, sampling valve rubber ring, etc. for contamination or damage at any time. If any,replace it in time.
